Alpha-Ketoglutarate (AKG)

Alpha-Ketoglutarate is a key metabolite in the Krebs cycle that declines with age. Supplementation with calcium AKG extended lifespan by 12% and compressed morbidity in mice (Buck Institute, 2020). It is involved in amino acid metabolism, collagen synthesis, and epigenetic regulation. Sold as the supplement Rejuvant (calcium AKG).

Mechanism of Action

AKG is a cofactor for alpha-ketoglutarate-dependent dioxygenases including TET enzymes (DNA demethylation) and Jumonji-domain histone demethylases, linking it to epigenetic regulation of aging. It inhibits ATP synthase and mTOR, mimicking aspects of caloric restriction. It also serves as a nitrogen scavenger, supports glutamate/glutamine metabolism, and is essential for collagen hydroxylation.

Dosing Research

Longevity dose: 1000-2000 mg/day of calcium alpha-ketoglutarate, taken on an empty stomach. The Rejuvant brand recommends 1000 mg/day. Some protocols use up to 3000 mg/day split into two doses. Clinical studies have used 1000 mg/day. Often combined with other longevity supplements.

Side Effects & Risks

Generally well-tolerated as it is an endogenous metabolite. Mild GI discomfort possible. Calcium AKG form provides calcium, which should be factored into total calcium intake. Very limited human longevity data. No significant adverse effects reported in available studies.
